    <p>Разработан эффективный подход к продукции практически нерастворимых гидрофобных белков в стабильной растворимой форме. Предложенный метод основан на получении слитых белков, включающих целевые белки и белок-носитель. В качестве носителя использован белок GrAD - апикальный домен термофильного шаперона GroEL. Для обеспечения возможности отделения белка-носителя путем избирательного гидролиза бромцианом был создан безметиониновый вариант GrAD. Изучены слитые белки с целевыми белками, соединенными с C-концом белка-носителя и слитый белок с целевым белком, соединенным с N-концом белка-носителя. Слитые белки экспрессированы в E. coli при различных температурах, они преимущественно образовывали тельца включения. Все слитые белки после растворения телец включения в растворе мочевины были успешно ренатурированы в стабильной растворимой форме. Изучена их стабильность и функциональность. GrAD как носитель может быть платформой для получения самых разных гидрофобных белков в растворимой форме для их исследования.</p>

   <trans-abstract xml:lang="en">
    <p>We have developed an approach to produce insoluble hydrophobic proteins in stable soluble forms. The proposed system is a fused protein construct, where a target protein is fused to the carrier. The carrier is GrAD protein, it is apical domain of thermophillic minichaperone GroEL. The Met-less version of GroEL apical domain has been made for further use of CNBr chemical cleavage. Studied proteins included hydrophobic insoluble proteins fused to the C- terminus of GrAD as well as its N-terminus. The fusions were expressed at various temperatures and inclusion bodies were harvested. All fusion proteins were dissolved in urea solution and successfully renaturated in stable soluble forms. Their stability and target protein functionality were tested. This approach can be used to solubilize and stabilize hydrophobic proteins for their research by different means.</p>
