Abstract (English):
A bioelectrochemical model for assessment of protease activity which based on detection of reduction currents of electroactivity compounds (toluidine blue), diffusing through gelatin gel, was come up. The electrochemical behavior of toluidine blue in gelatin gel is characterized by increase in the peak values of current in time and shift in potential of peak values of the current relative to the parameters obtained in the phosphate buffered saline. Introduction of trypsin in the electrochemical cell in concentration of 0,08 % (wt.) led to a change in nature of current-time relationship, which reflects a decrease in diffusion limitation. An indicator of protease catalysis in this model were the values of the reduction current and the rate of change in the current. The most significant changes in the current-potential curve were observed by the 10th minute of experiment. The development of the described model can be an electrochemical sensor of protease activity for the evaluation of enzyme preparations and cell cultures.
