IMMUNOCHEMILUMINESCENT ANALYSIS AS AN ALTERNATIVE TO THE TANDEMIC HPLC-MASSSPECTROMETRY METHOD FOR THE DETERMINATION OF VALPROID ACID

Abstract (English):
The work considers the immunochemiluminescent method, which acts as a reliable and more cost-effective alternative to a more modern method, such as HPLC-mass-spectrometry. Immunochemiluminescent method based on the complementary specific interaction of antigen and antibodies labeled with chemoluminescent labels. Such a labeling technique provides high accuracy and sensitivity of the method, allowing to detect very low amounts of the substance in which they are present in living organisms. The detection limit for the concentration of compounds is 2 mkg/ml. This becomes possible due to enzymatically enhanced chemiluminescence which occurs during the formation of 3-aminophthalate as a result of the reaction of luminol with a superoxide radical and the subsequent enhancement of chemiluminescence due to activators. This method does not lag behind analytical methods from more modern methods, but it does not have many of the negative features inherent in modern methods. The immunochemiluminescent method is relatively inexpensive, and the variability of analyzers that use this method allows it to be used in both large and small medical laboratories. The use of the immunochemiluminescent method in the ARCHITECT i2000 analyzer for determining valproic acid of children of different age showed the competitiveness of this method. The results obtained on the analyzer were characterized by high accuracy, sensitivity and specificity. Valproic acid values correlated with the age of children whose blood samples were taken.
